Just wondering, in the E3 trailer, he (she?) has a male voice with a Scottish accent.

But when he (she?) sees the giants, he (she?) lays an egg.

The egg laying is a common visual device to show fear, I wouldn't read so much into it is I were you... Jet Vac is male.

(lol imagine if Jet Vac was transgender... oh the parental outrage)

Yes, Jet Vac is a male. A lot of cartoons and animation use the egg laying to express that the character is in shock. It isn't only used with bird characters but other animals too such as mammals which don't even lay eggs. There is an episode (don't remember which, sorry) in Tom and Jerry were tom lays an egg when frightened (there are many other cartoons that do this but I can't remember all of them now. I think Courage the Cowardly Dog was another one).

But Yoshi from Mario will always remain a mystery to me…( I know that he's supposed to be a male but I want to say that “he’s” genderless like Quina from Final Fantasy 9 or Godzilla from the American Godzilla movie).

It's used to show a characters fear or surprise. Of course, you can't actually show someone ****ting themselves so substitutes are used instead; in this case an egg. Another example would be in Futurama, there is a running gag where Bender literally ****'s bricks.
