From the June edition of Game Informer Magazine:

If you are playing a two player game (which is once again limited to local play), the second player can also switch to a villain who isn't already in use.


My interpretation of that is that both players share access to the villain in the trap. Either can switch to him, but only if he isn't already in play.

Hmm... I've heard that all the villains are saved in the portal... Idk in which video but it said

I'll try to find this video again... So what I think is that the second player will choose a villain of the same element of the key before playing the game.

I play the game with my wife so it's not a deal breaker for me. The only thing I'd like is online PVP so I can actually use that feature...My wife isn't too good.
