I recently pre-ordered the new triple pack with whirlwind double trouble and drill sergeant and I also pre-ordered the Cynder single pack. I ordered them on the 7th and today I signed up for the amazon prime free trial to get them with free two-day shipping and I also changed my shipping preference from Group my shipments in to as few as possible to ship my items as they become available.
Will that change the date I was supposed to get them shipped? Does anyone know?

You will usually need to go into that order and manually change the shipping and grouping since you signed up for prime and changed your shipping preference after you had place that order.

They are not going to ship them earlier because you have prime but you will get them faster.

I know but will they send them later than they were originally before I got amazon prime?

Why would they do that? I've been using prime for years and it has no effect on when they send out your product unless you're ordering something that says it has "release date shipping" like video games and movies. It only affects how fast the shipping is.

My email estimates between January 27, 2012 - February 07, 2012.
Though, I ordered the triple character pack 6 and the character pack Cynder, they only gave an estimate for the 3 pack.

Just checked my order on the Amazon website, and there is already a different estimated date. At least it is an earlier date.

Just got my email for Cynder and Double Troble and it said the 17 of Jan to the30 of Jan.
